Slide 15. These are two adjacent slices from a whole breast. Each slice measures about 18 cm x 7 cm in area, and is 2 mm thick. After staining with hematoxylin, the slices are cleared in methyl salicylate and sealed in plastic bags, as shown here. Subcutaneous fat, mammary layer, retromammary fat and a portion of pectoralis major are seen. Each slice is examined at low magnifications (2-10 x, subgross level of observation) with a dissecting microscope and all lesions are located and numbered on the outside of the plastic bags with a wax pencil, as shown. The lesions are then cut out of the slices, photographed, and embedded in paraffin for routine histology. Hematoxylin stain.
